Identificación y cuantificación de biomarcadores en muestras de sangre de pacientes con enfermedades de depósito lisosomal (enfermedades raras) mediante técnicas analíticas avanzadas. (LISA)

Las enfermedades de depósito lisosomal (EDL) son un grupo de enfermedades raras que suponen un grave problema clínico, económico y social. Las técnicas actuales de medición de la actividad enzimática residual de las EDL no permiten diagnosticar y evaluar la progresión de las mismas. Por otro lado, las técnicas cromatográficas convencionales implican largos tiempos de análisis, el uso de un alto volumen de sangre y altos costes. El proyecto LISA, donde participan el CSIC (IP), La Fundación Española para el Estudio y Tratamiento de la enfermedad de Gaucher y otras lisosomales (FEETEG), la Universidad de Zaragoza y la empresa suiza CAMAG, tiene como objetivo principal la determinación de diferentes esfingolípidos considerados como biomarcadores de enfermedades de depósito lisosomal (EDL). Este objetivo se logrará mediante el desarrollo de: (i) una plataforma basada en tarjetas de muestra de sangre seca (DBS) acoplada a Cromatografía Líquida y Espectrometría de Masas (LC-MS) para llevar a cabo una rápida, cuantitativa y simultánea determinación de los diferentes liso-esfingolipidos con gran ahorro de muestra y mejores condiciones de extracción para los pacientes. Se utilizará una amplia colección de tarjetas DBS almacenadas en la Fundación FEETEG. (ii) Una plataforma automatizada basada en Cromatografía en Capa Fina de Alta Eficacia acoplada a Espectrometría de Masas (HPTLC-MS) que sirve de apoyo a la tecnología anterior.
